They do have a lift and her BM did go inside for a while. The lift only goes to the first floor though (main floor). It doesn't go upstairs (just bedrooms up there). So don't plan on a HM there if you or your FI is needs a wheelchair.
It was a great venue and may work for you. We had about 150 people in the Romantic Gardens for our ceremony. It holds 125 seated, so some folks stood. But our ceremony was only about 30 minutes and no one minded. There's also benches at the back and in the gazebos that flank the ceremony area. It is wheelchair accessible - my FIL is disabled and we had no trouble getting him around. They have wheelchairs to loan at the front desk and this worked out great for my grandmother, too. If you only do the ceremony there, I think it's pretty reasonable - around $1000.

Take a left (north) onto Josephine Street, go 3-1/2 blocks. Take a left into Denver Botanic Gardens parking lot between 9th and 10th Streets. The entrance to Denver Botanic Gardens is at 1005 York Street. Free parking is located across from the entrance.

People can be dropped off at the front, there is an elevator to the top - it holds tons of people, and you can have the ceremony there. It holds about 250 people and has an elevator. They also give you the ability to use their courtyard, or second floor club for a cocktail reception. It is a beautiful site.
The 1840s Ballroom, an elegant event site, features crystal chandeliers, custom-made velvet draperies, murals of historic Baltimore, an English pub bar, hardwood dance floors, and courtyard gardens. Accommodates 250 seated, 400 standing.

This is a large muesum with a lobby they rent out for weddings. It can be dark on foggy SF days. On a sunny day, it is pretty like in the photo. The whole museum is accessible, including the galleries by elevator.

The main ballroom holds 700 ppl and is very accessible. There are 2 more rooms upstairs that are for smaller groups (up to 140) which can be accessed by an elevator but the men's restroom is up a few steps.

It doesn't look it from this side of the blg, but this hall in the Oakland, CA hills is wheelchair accessible. It's very affordable to rent as well. By Calif standards anyway. It is not typically used for weddings and the acoustic equipment is not great. We had some difficulties with it. There is also no air conditioning, which is not usu an issue but on our day it was almost 100 deg! Since is is the site of priests' residences, music cannot be played in the hall after 10 PM.
